Knock Knock

Last night saw three couples, a cat, a dog and one young boy come together to watch the hockey. Well, everyone except the cat, who slept, and the boy (Linden) who just wanted to regale us with his best jokes:

Linden: Knock knock
Me: Who’s there?
Linden: Knock knock
Me: Knock knock who?
Linden: ….. (giggle).

Later:

Linden: Knock knock
Mr E Man: Who’s there?
Linden: Apple
Mr E Man: Apple who?
Linden: …………Pumpkin apple! (giggle giggle).

Another attempt:

Linden: Knock knock
Mr E Man: Who’s there?
Linden: Apple
Mr E Man: Apple who?
Linden: Running apple! (runs around the room).

A lesson from a pro:

Mr E Man: Knock knock
Linden: Who’s there?
Mr E Man: Orange
Linden: Orange who?
Mr E Man: Orange you glad I didn’t say apple?
Linden: blank look

After ordering pizza (Hawaiian with mushroom and vegetarian with chicken: that’s how we roll):

Linden: Knock knock
Me: Who’s there?
Linden: Mushroom
Me: Mushroom who?
Linden: Mushroom glad I didn’t say apple?

Methinks the boy needs some more lessons.

hockey hockey HOCKEY!

Season opener tonight!

I was planning to write a nice long, thoughtful post about how this life-long football and rugby fan got into hockey after arriving in Vancouver. I was going to tie my experiences in to musings on the very essence and nature of sport, and contrast the national characters of my two favourite countries.

But my looming grant deadlines prevent me from writing anything more profound than the following:

Signs of the times

With just one week to go before the Canadian general election, Vancouver lawns are bearing fresh crops of political signs. My ride to work is flanked with splashes of red, blue*, orange, and occasionally green. One house down our street even has Liberal, NDP and Conservative signs, side by side on the front lawn, which I don’t understand. Not even one little bit, unless it’s intended as a joke.

But then again I’ve never really seen the point of having even one sign on your property. Are people really influenced by indicators of how their neighbours are voting? Might it sway them into voting tactically, say if they get scared by the number of Conservative party signs?

Every time I see one of these things, I think about how easy it would be to turn the text into “Salmon RAPER”. One good brush full each of white and of blue paint… Just sayin’. (I absolutely do not condone acts of rebellion against the fascists vandalism. Besides, I’m too scared to break the law before I get citizenship).

Looks like someone else had no such qualms…

But aren’t the parties who are doing the worst in the polls going to be the most aggressive about putting signs in their supporters’ gardens? That certainly seems to be the case in my riding, where those nasty blue signs are everywhere but the Conservatives are apparently a distant third in the polls behind the NDP and Liberals, and might even finish behind the Greens.

What really doesn’t make sense is the house I pass every day that has an Obama-Biden sign in the window. Dude, this is Canada, FFS.

*Note to American readers: in Canada, the UK, and every other country that I can think of except for the US, the left wing party uses red and the right wing party uses blue. If anyone knows the history behind the decision to use the opposite colour scheme in the US, please let me know! It’s something that bugs me and no-one seems to know the answer…

NO SOUP FOR YOU!!!

The soup recipes are up. All three recipes look amazing, but Dr Isis gets my vote this week for having two fabulous recipes, introducing the soup swap idea, and making me splurt out my tea as I laughed at the caption to Figure 1.

Unfortunately I don’t get to play this time, because Mr E Man is the soup maker in this marriage (his family all have awesome soup genes, his Mum’s French Onion is to die for) and he refuses to divulge his secrets to the internet. It’s soooo unfair, he is such a meanie. His best soups are

a) cauliflower, leek and blue cheese, and
b) butternut squash, carrot and ginger,

but I have no idea how to make them. Sorry.

Blast from the past

Isn’t high throughput science cool?
Wanna find out which host proteins are required for HIV infection? Do a whole genome screen! Try to infect human cells with HIV while using short interfering RNAs to individually knock down every single one of their genes, and see which ones become resistant to infection. Voila, 237 novel proteins / potential drug targets. Easy peasy.
Maybe that bores you, and you’d rather map all the genomic rearrangements that occur in human lung cancer cells. No problem at all, just sequence everything in sight and find all 409 rearrangements, down to the single base pair level. Done! Now let’s go for lunch.
I’m being flippant, of course – these studies require a massive amount of preparation, careful experimental work and especially data analysis, and I have nothing but respect for the researchers who are extracting the maximum benefit from these new technologies. It’s just a totally different ball game to my own days in the lab, where most papers were still of the one-gene-at-a-time variety.
So I was thrilled to spot an old-school paper recently that is closely related to my PhD thesis work. I worked on the v-Jun protein, a copy of a cellular regulatory gene that was picked up and mutated by an avian retrovirus, and which causes sarcoma formation when introduced back into chicken cells. While my lab was interested in comparing the mechanisms of transcriptional and cell growth regulation by the cellular and viral versions of the gene, most papers that were published in our field at the time reported the discovery of a new transcriptional target of v-Jun.
The paper, published by a lab with whom I corresponded during my PhD work, describes the regulation of the TOJ3 gene by v-Jun. The original paper describing the upregulation of TOJ3 transcripts in v-Jun transfected cells came out in 2001, the year I started to write up my thesis. I was therefore familiar with the work, but hadn’t seen any related papers published in the intervening years. I have no idea why this follow-up paper took seven years to come out, but I’m glad it finally got published – reading the introduction and methods sections rendered me all nostalgic. CAT assays! I didn’t think anyone did those any more. (They were renamed CATH assays in my old lab after I did about three million of them in my final year in the lab). And I was delighted to discover that I can still spot a consensus Jun binding site at twenty paces (TGA G/C TCA, since you ask. A palindrome, which I always think is cool).
Anyway. It is now confirmed that TOJ3 is a direct transcriptional target of v-Jun. A nice solid piece of research, and a wonderful blast from the past for me.

Poor planning or Conservative conspiracy?

I’ve just realised that the English language debate between the five main Canadian party leaders is going to clash with the US Vice Presidential debate. Damnit! I originally thought the clash was with the French language debate.

What’s an effectively unilingual, voteless but interested, Canadian resident to do? I really want to see what my next prime minister has to say for himself. But on the other hand I really really want to see Biden make Palin break down into incoherent babbling on live TV. Hmmm.

Well, since we only have one TV, I guess it comes down to how badly Mr E Man wants to see the Canadian debate. He already knows who he’s voting for, so hopefully I can persuade him to watch the Palin train wreck and either record the Canadian debate or just watch all the analysis shows afterwards.

If I didn’t know any better I’d say Stephen Harper had deliberately called the election at a time when it was bound to be overshadowed by media coverage of the American Presidential race. But surely he wouldn’t do such a thing…
